(A) Flow cytometry was used to identify the percentage of CD11b + F4/80 + macrophages within the CD45 + population of the mammary gland during development. Gating based on fluorescence minus one (FMO) controls is shown. (B) FACS analysis of wild-type and Ackr2 −/− -deficient glands was carried out at 6, 6.5, 8 and 12 weeks. Results were combined from two independent experiments, n =8-11. (C) <t>Mac2+</t> cells, indicated by arrowheads, were visualised within the developing mammary gland by immunohistochemistry. Representative 40× bright-field images of Mac2-stained sections from wild-type and Ackr2 −/− glands are shown.
